Abstract
Dryopteris crassirhizoma dryocrassin ABBA is a kind of phloroglucinol compound isolated from the whole plants of Dryopteris crassirhizoma. While previous studies have shown that dryocrassin ABBA induces various defence responses in potato tubers, limited information is known about the mechanism of dryocrassin ABBA-induced plant responses. The transcriptome profiles in potato tubers after treatment with dryocrassin ABBA or distilled water were analysed using RNA-Seq. The profiling analysis revealed a total of 37 million clean reads and 3252 differentially expressed genes (DEGs). The data were further confirmed by real-time PCR. The results suggest that dryocrassin ABBA treatment enhances resistance in potatoes, which may be primarily mediated by the salicylic acid (SA) signal pathway, while the jasmonic acid (JA), the ethylene (ET) and the abscisic acid (ABA) signal pathways effectively induce resistance in potatoes. Moreover, the transcripts related to flavonoid metabolism were also up-regulated in potatoes after dryocrassin ABBA treatment, demonstrating that secondary metabolites are related to the constitutive activation of the defence response in dryocrassin ABBA-treated potatoes. These findings reveal that the induced resistance of dryocrassin ABBA is related tomultiple signaling pathways and defence components to enhance host defence responses in potato tubers.
Resumen
Dryopteris crassirhizoma dryocrassin ABBA es un compuesto tipo floroglucinol aislado de las plantas completas de Dryopteris crassirhizoma. Mientras que estudios previos han mostrado que dryocrassin ABBA induce diversas respuestas de defensa en tubérculos de papa, se tiene conocimiento de información limitada acerca del mecanismo de las respuestas de las plantas inducidas por dryocrassin ABBA. Se analizaron los perfiles del transcriptoma en tubérculos de papa después del tratamiento con dryocrassin ABBA o agua destilada utilizando RNA-Seq. Los análisis de perfiles revelaron un total de 37 millones de lecturas claras y 3252 genes expresados diferencialmente (DEG’s). Los datos se confirmaron aun mas por PCR de tiempo real. Los resultados sugieren que el tratamiento con dryocrassin ABBA aumenta la resistencia en papas, lo cual pudiera mediarse primeramente por la ruta de señales del ácido salicílico (SA), mientras que las rutas de señales del ácido jasmónico (JA), el etileno (ET) y el ácido abscísico (ABA), efectivamente inducen resistencia en papa. Mas aun, los transcriptos relacionados al metabolismo de los flavonoides también fue regulado en papas después del tratamiento con dryocrassin ABBA, demostrando que los metabolitos secundarios están relacionados a la activación constructiva de la respuesta de defensa en papas tratadas con dryocrassin ABBA. Estos descubrimientos revelan que la resistencia inducida de dryocrassin ABBA esta relacionada a múltiples rutas de señalización y componentes de defensa para aumentar las respuestas de defensa del hospedante en tubérculos de papa.
