Relative transcript levels and standard deviations (SD) were determined from the relative standard curve constructed from cDNA dilutions and divided by the target quantity of the calibrator (Pagano et al., 2002). The specificity of the qRT-PCR products was proven by appropriate melting curves (specific melting temperature) and by the expected size of the PCR products, determined by gel electrophoresis.

Different cell lines were used: S1.1, S6, S7, M and M2 cells are SKNBE2 neuroblastoma cells (provided by the cell bank of the National Institute of Cancer Research (IST) Genoa, Italy and obtained from ECACC, [19]) stably transfected with pNDM29-EGFPN1 or pEGFPN1 empty vector respectively as described in [20] (both grown in RPMI supplemented with 10% FBS, 1% glutamine and 200 μg/mL G418) and transiently transfected with pNDM29-EGFPN1, pAntiNDM29-EGFPN1 (pS1.1/A) and pEGFPN1 (grown in DMEM supplemented with 10% FBS, 1% glutamine). The v-myc-induced Q83 lipocalin is a siderocalin
2010, Journal of Biological ChemistryCitation Excerpt :Despite the fact that Q83 and NGAL share limited sequence similarities, they are clearly homologous proteins, as both proteins have surprisingly similar ligand binding affinities and display highly conserved ligand binding modes. In addition, similarly to NGAL, Q83 and the chicken homologue Ch21 seem to be involved in inflammation (11, 12, 55, 56), tissue differentiation (13, 28), and cancer progression (17, 27). These similarities strongly suggest that Q83 and NGAL can be considered to represent siderocalin family members with comparable biological functionalities and emphasize the presumed pleitotropic functions of siderocalins.
