The Staudinger reduction provides a bio‐orthogonal and generalizable approach to small‐molecule‐triggered protein activation. We have developed three genetically encoded caged lysine analogues that can be rapidly activated with phosphines in order to control protein function in mammalian cells. This was applied to the conditional control of protein translocation and triggering SUMOylation, an important post‐translational modification, of a caged target protein.
