Molecular barcoding is used to tag nucleic acids with sequence-encoded identifiers to distinguish between different samples or individual molecules. I describe a one-pot procedure for efficiently tagging linear dsDNA with molecular barcodes. This approach has potential utility for high-throughput in vitro biochemical experiments that require uniform and quantitative dsDNA tagging.
