Cyclotides are natural occurring microproteins found in plants that are stabilized by three disulfide bonds and a head-to-tail circular backbone forming a cyclic cystine-knot (CCK) motif. Here, the suite of cyclotides obtained from butterfly pea plant (Clitoria ternatea) is described using a diagnostic method that rapidly classifies the sequences as Möbius or Bracelet subtypes on the basis of mass-spectral product ion fingerprints caused by the Xxx-Pro bond fragmentation in complex, natural, microheterogeneous peptide libraries.
