The Taxonomic Validity and Phylogenetic Relationships of Genus Platycephalus Bloch, 1785 (Teleostei: Platycephalidae) in the Northwest Pacific Inferred from the Mitochondrial Genome

Genus Platycephalus in the Indo-West Pacific comprises about 20 species, of which only five are known in the northwest Pacific. In this study, we sequenced and annotated the complete mitogenome of 4 species: Platycephalus indicus, Platycephalus cultellatus, Platycephalus sp., Platycephalus sp. 2, and downloaded two complete mitogenome of Platycephalus from GenBank for comparative mitogenome researches. The complete mitogenomes of the 5 species (Platycephalus indicus, Platycephalus cultellatus, Platycephalus sp., Platycephalus sp. 1, Platycephalus sp. 2.) are 16,560, 16,672, 16,548, 16,552, and 16,544 bp in length respectively. The mitogenomes are similar in organization and structure with usual mitogenomes of fish species, containing 13 protein-coding genes (PCGs), 22 transfer RNA genes, 2 ribosomal RNA genes, and 2 non-coding regions (control region and L-strand replication origin, OL). The analysis of genetic distances shows that the variations between 5 Platycephalus species are 0.051 ~ 0.160 on mitogenomes, 0.060 ~ 0.185 on PCGs, and 0.024 ~ 0.085 on rRNA genes, supporting the taxonomic validity of Platycephalus sp., Platycephalus sp. 1, Platycephalus sp. 2. Phylogenetic relationships of genus Platycephalus were first carried out based on mitogenome. The results reveal that Platycephalus indicus was split at the basal clade. Platycephalus sp. and Platycephalus sp. 2 were the most closely clustering with Platycephalus sp. 1 and next with Platycephalus cultellatus. The distribution, relationships, and evolution history of Platycephalus were discussed to provide basic information for taxonomy and phylogenetic relationships of genus Platycephalus.
