Chimeric molecules comprising an Xrn1-resistant RNA motif (xrRNA) and a small interfering RNA (siRNA) duplex were engineered. The molecules self-assemble in vitro, resist degradation by the conserved eukaryotic 5’ to 3’ exoribonuclease Xrn1, and trigger gene silencing in mammalian cells. This work sets the stage for further investigations into using xrRNA motifs as building blocks in RNA-based nanotechnology.
