Dynamic constitutional chemistry is an extremely powerful tool to control the activity of Carbonic Anhydrase (CA) in the presence of dynamic libraries. These adaptive systems operate within the inner CA active site or on the outer enzyme microenvironment used to overexpress specific dynamic frameworks that lead to the regulation of CA activity. This review describes incipient examples toward the generation of chemical diversity and their functional amplification toward effective strategies for CA inhibition and activation.
