Microfabricated Probiotic Formulation for Inhibition of Salmonella

Probiotics are considered a promising alternative to antibiotics for Salmonella disease control. However, probiotics fed to poultry and humans lose most of their activity in the stomach because of their sensitivity to gastric fluids. For this purpose, we developed a universal platform for probiotics, shaped as micrometer-scale conical frustums based on microfabrication technology. The casted probiotics produced through molding/demolding processes were coated with a protection layer made of an anionic copolymer, Eudragit S 100. The samples exhibited a gradual release in a simulated gastric fluid (SGF), followed by a rapid stepwise release upon exposure to a simulated intestine fluid (SIF). The encapsulation, viability, and environmental stability were characterized by using Lactobacillus acidophilus (L. acidophilus) encapsulated probiotic formulations. The microfabricated probiotic patch provided protection to encapsulated L. acidophilus cells against harsh digestive conditions and environmental storage conditions. This study showed that microfabricated probiotic formulations can enhance the efficacy of probiotics and result in a dose-sparing effect, reducing the required amount of cells and costs.
