Antioxidant Capacities, Antimicrobial Activity, Phenolic Contents and α-Amylase Inhibitory of Salvia leriifolia Extracts from Sabzevar

One of the major issues in the control of type 2 diabetes is to reduce hyperglycemia after sugar intake, which is possible through inhibition of enzymes such as α-amylase. In this study, we decided to investigate the inhibitory effect of plant extracts of Salvia leriifolia (Norouzak) on bacterial α-amylase and pancreatic enzymes. The results indicated that hexan and methanol extracts of S. leriifolia had the strongest α-amylase inhibitory effect with IC50 values of 25.6 and 30.5 mg/ml, respectively. The results exhibited a competitive inhibition kinetics towards starch as a substrate for both α-amylases. The fluorescence spectrum of the enzyme revealed that the enzyme structure was opened in the presence of herb extracts. Using the DPPH assay, antioxidant activity was also measured. Antibacterial effect of plant extract was tested against three bacteria, Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli; furthermore, MIC and MBC values were determined for each bacteria by the microdilution broth method. The maximum inhibitory effect was observed in extracts of the bacterium P. aeruginosa with MIC = 80 mg/ml. Folin–Siocalcho reagent was used to measure the phenol of extract, and the amount of flavonoids was determined based on colorimetric aluminum chloride method. Statistical analysis using SPSS depicted that the phenol and flavonoid contents were significantly correlated with the antioxidant activity of the plant. According to the amount of IC50 compared with synthetic antioxidants, Norouzak is suitable for using as treatment or a natural preservative to replace the synthetic variety.
