Tomentosin attenuates behavior deficits and neuroinflammatory response in MPTP-induced Parkinson’s disease in mice

Parkinson’s disease (PD) is the second most neurodegenerative disease, which affects many persons worldwide. Tomentosin is an active compound isolated from I.viscosa plant that has extensive therapeutic values. In this exploration, the neuroprotective actions of tomentosin were investigated against the MPTP-stimulated neuroinflammation in mice. The PD was stimulated to the C57/BL6 mice by injecting the 20mg/kg of MPTP for 4 times with a 2h interval for 15 days along with the tomentosin treatment. The Rota-rod test, grasping test, and pole climbing test was executed to investigate the motor functions of test animals. The statuses of pro-inflammatory cytokines, reactive oxygen species, and myeloperoxidase were examined by using the commercial ELISA kits. The superoxide dismutase enzyme level was inspected by the standard method. The expression level of TLR-4/NF-ƙB was examined by western blotting analysis. The brain tissues of investigational animals were analyzed microscopically. The tomentosin treatment to the MPTP-intoxicated PD mice was displayed an appreciable regain in the bodyweight. Tomentosin notably prevented the MPTP-stimulated impairments in the motor functions. The status of pro-inflammatory cytokines, ROS, and MPO was assuaged by the tomentosin treatment in the brain tissues of PD mice. The tomentosin inhibits the TLR-4/NF-ƙB signaling pathway and also prevented the inflammation-mediated neuronal cells damages in the brain tissues of PD animals. The reduced glial cells damages and usual ganglion layers were seen in the brain tissues of tomentosin supplemented PD mice.
