Cyclotides are a fascinating class of macrocyclic plant peptides that contain a cyclic cystine knot, a compact and highly stable structural motif. This review by D. J. Craik and co‐authors is an historical overview of the developments in NMR spectroscopy, as well as X‐ray crystallography, that have enabled the resolution of cyclotide structures and conclusive determination of disulfide connectivities of several exemplar cyclotides. Such work has been inspired by the innovative and pioneering studies of Professor Horst Kessler in cyclic peptide chemistry and NMR spectroscopy. Cover art by P.J.H.
