In the present study, we analyzed and identified two CXC chemokine ligands in the swamp eel, Monopterus albus. The open reading frames of MaCXCL13 and MaCXCL14 were 375 bp and 300 bp, which encoded putative proteins of 124 aa and 99 aa, respectively. Phylogenetic and protein similarity analyses found that only MaCXCL14 has high homology with CXCL proteins from other vertebrates, while MaCXCL13 shared low homology with those of mammals. Tissue expression and in vitro expression analysis revealed different expression patterns of these MaCXC chemokines. MaCXCL13 was mainly expressed in immune tissues, such as the spleen and kidney, whereas MaCXCL14 was highly expressed in the brain, followed by the spleen. Upon induction with polyinosine-polycytidylic acid, the transcription levels of the two CXC chemokine genes were upregulated, while phorbol 12-myristate 13-acetate (PMA) only induced the upregulation of MaCXCL13. MaCXCL14 expression was downregulated after PMA stimulation. These results suggested that the two MaCXC chemokine ligands are associated with the immune response and homeostatic processes in the swamp eel.
