Depth-resolved imaging of intercellular structures of large intestine with polarization-sensitive FFOCT

Abstract A polarization-sensitive full-field optical coherence tomography (PS-FFOCT) system was developed, which is capable of revealing both the reflectivity and phase-retardation caused by the anisotropic samples simultaneously with the lateral resolution of 0.98 µm and the axial resolution of 2.4 µm. The system was used to generate depth-resolved en face isotropic intensity images and birefringent images of the normal and abnormal large intestine tissue samples of human in vitro. To our knowledge, for the first time, the intestine gland cells and intercellular structures of normal intestine and infiltration of metaplasia of cancerous intestine tissue were observed in the en face images obtained with PS-FFOCT device. The resolution and effects of measurement errors are discussed. The results obtained in this work demonstrated the potential applications of PS-FFOCT technique in the rapid intra-operation diagnosis of cancers.
