A reprogramming platform: An enzyme‐linked immunosorbent assay (ELISA) using a combination of small‐molecule probes can detect subtle structural changes in the substrate binding pockets of aryl acid adenylation (A) domains. Combined with a directed‐evolution approach, the ELISA technology can be used to isolate aryl acid A‐domains with the desired activity.
