Anticancer activities of Careya arborea Roxb on MCF-7 human breast cancer cells

Careya arborea (CA) is a plant found in Thailand and consumed daily as a fresh vegetable. This study evaluated the effects of treatment with CA against human breast cancer, MCF-7 cell lines. Effects of ethanolic extract of CA leaves were measured on cell viability, migration and its mechanism. Methods were performed by sulforhodamine B (SRB), colony formation, wound healing, gelatin zymography, RT-PCR and Western blotting assay. The result indicated that CA inhibited MCF-7 cancer cell viability in a concentration-dependent manner with very low IC50 values of 34.78 ± 1.30 and 18.88 ± 2.18 μg/mL after treatment with 24 and 48 h, respectively. CA inhibited colony formation with a lower IC50 value of 3.48 ± 0.15 μg/mL than found from cell death. The studies showed that CA leaf extracts enhanced the process of apoptosis by inducing significant ROS generation, increasing caspase 3 activity and consistently reducing mitochondrial function. Additionally, CA leaf extracts significantly reduced cyclin D1 protein along with increasing caspase 3 and cytochrome c protein. Interestingly, low concentrations CA leaf extract (5 μg/mL) induced greater apoptotic bodies and cell shrinking compared to the untreated control groups. Finally, CA leaf extract potently suppressed the migration of MCF-7 cells accompanied by downregulation of the MMP 2 and MMP 9 protein expression. These results indicate that CA leaf extract has an anticancer activity against MCF-7 cells and is maybe useful for breast cancer prevention and therapy.
