Isolation and characterization of Alternaria GFAV15, an endophytic fungus from green fruit of Tinospora cordifolia (Willd.) Miers from semi-arid region

Abstract An endophytic fungus, Alternaria GFAV15 with antimicrobial potential was isolated from the green unripe fruit of Tinospora cordifolia, a well-known medicinal plant of semi-arid region of Rajasthan, India. The isolated endophytic fungus from green fruit has been identified on the basis of cultural and morphological characters and was characterized by internal transcribed spacer (ITS 1 and ITS 4) sequence analysis. The fungus sequence has been submitted at NCBI with accession number MK212342 with the name Alternaria GFAV15. Thin-layer chromatography (TLC) guided bioautography was performed to localize and unlock the antimicrobial bioactive compounds within the isolated fungal extract. Methyl esters (hexadecanoic acid and octadecanoic acid) were identified and recorded by GC-MS (gas chromatography-mass spectrometry) as the major compounds in active sub-fraction (RF 0.257). These compounds showed strong antibacterial activity in combination against Staphylococcus aureus (Gram positive) and Escherichia coli (Gram negative).
