Rule breakers: The trans‐acting adenylation domains Cal22Thr and Cal23Asn are shown through gene disruption experiments and in vitro assays with purified enzymes to play essential roles in the biosynthesis of the nonribosomal peptide WS92326A in S. calvus. For soluble heterologous protein production, Cal22Thr and Cal23Asn require coexpression with the MbtH‐like protein Cal4, the latter copurifying with Cal23Asn as a complex.
