Photosynthesis-related protein expression in sugarcane during sugarcane– Fusarium verticillioides interaction revealed by iTRAQ

Pokkah boeng disease (PBD) is mainly caused by Fusarium verticillioides and F. proliferatum, and it substantially reduces sugarcane yield and quality in China. Sugarcane proteins play important roles in defense against these fungal infections. This study aimed to explore the photosynthesis-related proteins involved in PBD resistance in resistant (R) and susceptible (S) sugarcane materials. The main photosynthetic parameters were measured with the LI-6400 photosynthesis system, fluorescence measurements were acquired via saturation pulse, and an isobaric tags for relative and absolute quantification (iTRAQ) and low PH nano-HPLC–MS–MS-based proteomic techniques were used to assess photosynthesis-related proteome changes 14 days after inoculation with F. verticillioides between inoculated and control leaves of YT94-128 (R) and GT37 (S) plants. The net photosynthetic rate (Pn), stomatal conductance (gs), and transpiration rate (E) of sugarcane plants were significantly reduced as pathogen stress increased, and the intercellular CO2 concentration (Ci) was significantly elevated compared to that in the control plants. In addition, 29 proteins involved in photosynthesis were found to be differentially expressed following pathogen inoculation, including six proteins that were differentially expressed in both sugarcane varieties. These results indicate that complex photosynthesis-related proteins are involved in the sugarcane–F. verticillioides interaction and provide novel insights into the molecular mechanisms underlying sugarcane response to PBD pathogen infection.
