The development of novel bioconjugation strategies is useful for various therapeutic and diagnostic applications. This work describes the synthesis and genetic encoding of an unnatural amino acid that serves as a functional handle for numerous bioconjugations. In particular it can be employed in bioorthogonal [2+2+2] cycloaddition reactions to either conjugate a small‐molecule probe/drug or to immobilize a protein on a surface.
