Development of a naphthlimide-based fluorescent probe for imaging monoamine oxidase a in living cells and zebrafish

In this work, we have developed a ratiometric fluorescent probe (named Probe A) for the detection of monoamine oxidase A (MAO-A) in living cells and zebrafish. The newly developed Probe A is an analogue of Probe 1 in a published research (Anal. Chem. 2016, 88, 1440), and the only structural difference between the two probes is the length of the alkyl group connected to the nitrogen atom of the 1,8-naphthalimide fluorophore. By chance, we have found a convenient synthetic method of 4-hydroxyl-1, 8-naphthalimide derivatives, making the synthesis of Probe A and Probe 1 much easier. Experimental data shows that the ratio of fluorescence intensity at 550 nm and 452 nm (I550/I452) changed 186-fold after the reaction triggered by MAO-A. Probe A was more reactive than Probe 1 and much less affected by temperature in the detection of MAO-A. Furthermore, Probe A was successfully applied to imaging endogenous MAO-A in living cells and zebrafish by confocal laser scanning microscope.
