Expanding on CRISPR/Cas systems : We describe a chemo‐enzymatic method for synthesizing chemically modified sgRNAs that bear a 3′‐biotin functional group. The biotinylated sgRNA was used as an affinity reagent to discover endogenous human proteins capable of binding the sgRNA.
