Abstract
During wound healing, bacterial infection and elevated reactive oxygen species (ROS) levels often lead to de layed healing and potential pathological complications. Traditional wound dressings are difficult to have the ef fects of sustained anti-bacteria, scavenging ROS and promotion of angiogenesis simultaneously. This study inno vatively constructed a novel electrospun nanofibrous membrane (NFMs) composite as wound dressings to ad dress delayed healing caused by bacterial infections and ROS-rich microenvironments in chronic wounds. The novel NFMs wound dressing (Ga-CurNPs@COF-NFMs) was composed of gallium-curcumin nanoparticles (Ga CurNPs) encapsulated by covalent-organic framework (COF). The Ga-CurNPs show good antibacterial and an tioxidant capabilities and the high porosity of NFMs and COF ensures controlled release of Ga-CurNPs. The physicochemical properties, release characteristics, biocompatibility, antibacterial properties, anti-inflammatory and in vivo retention of Ga-CurNPs@COF-NFMs were evaluated. At the cellular level, the pro-angiogenic and an tioxidant effects of Ga-CurNPs@COF-NFMs and their mechanisms were explored. Finally, the effect of Ga CurNPs@COF-NFMs on wound healing in rats was investigated, and the in vivo antibacterial mechanism was ex plored. This study not only provides a novel strategy for chronic wound treatment but also contributes to the de velopment and promotion of chronic wound therapy.
