We developed a label-free method to measure sortase activity by using transpeptidation-directed intramolecular bipartite tetracysteine display. This assay is homogenous with a detection limit of 10.7 ng mL⁻¹, and it can be applied to characterize Sortase A (SrtA)-targeted inhibitors, providing a new approach for anti-virulence drug discovery.
