The cover feature picture shows the in vivo generation of cyclic peptides through the protein‐splicing reaction carried out by split‐inteins. With this method, cyclic peptide libraries can be genetically encoded, expressed and used in screening campaigns. Here we show that the concentration of the intracellular cyclic peptide not only strongly depends on the split‐intein used, but also on the target sequence. Crystal structures of the pre‐splice complexes of the inteins provide further insights into their molecular mechanisms. More information can be found in the communication by S.
