We designed, constructed, and validated a phage-displayed single-framework synthetic antigen-binding fragment (Fab) library based on a modified trastuzumab framework. High affinity and specificity for human epidermal growth factor receptor-3 (HER3) was shown by an anti-HER3 Fab isolated from the library. It selectively accumulated in a murine HER3-postive xenograft, thus providing a lead for the development of HER3 imaging probes.
