New paradigm for potency: The crystal structures of E. coli IspH in complex with (E)-4-mercapto-3-methylbut-2-en- 1-yl diphosphate and (E)-4-amino-3- methylbut-2-en-1-yl diphosphate enabled us to design IspH ligands with improved binding capacities. A novel binding feature gave rise to compounds that present a new paradigm for the development of more potent IspH inhibitors.
